Obtaining of Pathogen - free Stock and Clonal Multiplication through the Carnation Shoot Apex Culture in vitro


Abstract
This study was performed to obtain of virue-free stock and multiplication through the shoot apex culture in vitro with angel carnation (cv. Scarlet bell). The primary liquid static culture was examined by comparing various growth regulators, pHs and storage duration of cuttings for their effect on growth and development of multiple shoots from shoot apices. When shoot apex (about 0.2§® height) with two leaf primordia and apical meristem of dome shape cultuzed on liquid static culture in Murashige and Skoog (MS) medium with NAA and BA of various concentration, apical dominance was losed and morphologically normal multiple shoots were proliferated rapidly in axil of leaf, there after have been keeping normal development. And then multiple shoots were separated one by one for the purpose of multiplication and subcultured in MS solid medium (0.7% agar) without growth regulators. After a month transferred subculture was made to multipleshoot with 15.8 shoot over from a shoot and rooted plantlets were formed 80.5 percent.
